A newly emerging variant of Influenza A (FluA) H3N2, referred to as Subclade K, formerly J.2.4.1, has been increasing in prevalence in the Northern Hemisphere since mid-2025. 1, 2

In-silico alignment confirms that our EasyScreen™ Respiratory Pathogen Detection Kit (RP007) will efficiently detect all currently known sequences of the FluA H3N2 Subclade K variant.

Our unique approach:

  • Pan-Influenza A targeting of the Matrix Protein gene, avoiding the highly variable Hemagglutinin (H) and Neuraminidase (N) regions.
  • Powered by 3base® technology, the assay is more resilient to common mutations, ensuring robust detection of emerging variants. 3

 

In-silico Assessment of Influenza A Subclade K

A total of 330 Influenza A H3N2 clade 3C.2a1b.2a.2a.3a.1 sequences collected in the United Kingdom between 01 August 2025 and 18 November 2025 were obtained from GISAID 4 and aligned with the EasyScreen™ Respiratory Pathogen Detection Kit (RP007) following 3base® conversion.

  • 317 sequences (96%) showed a perfect (100%) match.
  • 13 sequences (4%) contained a single mutation mismatch in one primer region. The location of these mismatches is not expected to impact the assay’s capability of detecting the Influenza Subclade K variants.

Therefore, 100% of the analysed sequences of the emerging Subclade K variant remain within the expected detection range of Genetic Signatures’ EasyScreen™ Respiratory Pathogen Detection Kit.
